传统细胞培养是在单层中生长,细胞是扁平并可以在水平面上自由粘附和扩散,但不支持在垂直方向上扩散;2D表面上培养的细胞具有强烈的顶端-基底极性,这种极性与某些细胞类型相关,例如上皮细胞;3D细胞培养即三维细胞培养,是指具有三维结构的不同材料载体与各种细胞在体外共培养,使细胞在载体的三维立体空间结构中迁移,生长,形成三维的细胞-载体复合体,主要用于细胞球,类器官等培养。
作为疾病验证模型,相比于传统2D疾病模型,以3D为基础培养的类器官在阐明疾病的发展,稳态性和发病机理等与体内环境更加接近,在多领域将发挥重要,诸如细胞治疗,药物研发,基因工程,免疫研究,组织再生等领域。基质胶作为类器官3D培养的重要载体,在其中发挥着举足轻重的作用。

基质胶是从富含胞外基质蛋白EHS小鼠肿瘤中提取中的可溶性基底膜制备物,其主要成分由层粘连蛋白,IV型胶原,硫酸乙酰肝素蛋白聚糖(HSPG)和巢蛋白组成,还包含生长因子如TGF-beta,EGF,IGF,FGF,组织纤溶酶原激活物和EHS肿瘤自身含有的其他生长因子,为组织和细胞提供支撑,抗拉强度和支架支撑,也可以作为细胞粘附和运动的三维亚结构以及生长因子,趋化因子和细胞因子的储存库,同时可作为细胞形态发生和分化的信号等。
